Breaking the 3-D Diffraction Limit

"Super-resolution" fluorescence microscopy has allowed researchers to break the diffraction limit of visible light and achieve near-molecular image resolutions -- in two spatial dimensions. Huang et al. pushed that advantage into 3D, through a stochastic approach that takes advantage of optical astigmatism to determine both the axial and lateral positions of individual molecules with nanometer accuracy. In this movie, they show a 3D "STORM" image of five microtubule filaments from a monkey kidney cell; the scale bar is 200 nanometers in length.
